Description

DUOXA2 encodes a maturation factor essential for the functional expression of dual oxidase 2 (DUOX2). DUOXA2 facilitates the translocation of DUOX2 from the endoplasmic reticulum to the plasma membrane and is required for DUOX2 enzymatic activity, which generates hydrogen peroxide (H2O2) used in thyroid hormone synthesis. Mutations in DUOXA2 cause congenital hypothyroidism due to defective H2O2 production.

Mutation functional classification

Loss of Function (LOF)

Most reported DUOXA2 mutations are loss-of-function, leading to reduced H2O2 production and congenital hypothyroidism.

Gain of Function (GOF)

No gain-of-function mutations have been reported.

Dominant Negative (DN)

No dominant-negative effects have been described.

Pathways

• Thyroid hormone synthesis (KEGG hsa04918)
• NADPH oxidase activity

Protein Summary

DUOXA2 is a 320-amino acid transmembrane protein localized to the endoplasmic reticulum and plasma membrane. It acts as a chaperone for DUOX2, ensuring its proper folding and transport. Without DUOXA2, DUOX2 remains inactive and retained in the ER, leading to deficient H2O2 production.
