Description

DRG2 (Developmentally Regulated GTP-Binding Protein 2) is a protein-coding gene located on chromosome 17q25.1. It encodes a member of the DRG family of GTP-binding proteins, which are evolutionarily conserved and involved in cell growth, differentiation, and stress responses. DRG2 is ubiquitously expressed and plays roles in translation regulation, cell cycle control, and apoptosis. Its dysregulation has been implicated in various cancers and neurodevelopmental disorders.

Loss of Function (LOF)

Missense mutations (e.g., p.Arg116His, p.Arg168Trp) impair GTP binding and reduce cell proliferation.

Gain of Function (GOF)

Not reported in curated databases.

Dominant Negative (DN)

Not reported in curated databases.

Pathways

• GTPase signaling (Reactome: R-HSA-9012999)
• Translation regulation (KEGG: hsa03013)

Protein Summary

DRG2 is a 364-amino-acid GTP-binding protein with a conserved G domain. It functions as a molecular switch, cycling between GTP-bound (active) and GDP-bound (inactive) states. DRG2 interacts with ribosomal proteins and translation factors, modulating protein synthesis. It also participates in stress granule formation and cell survival pathways. Structural studies reveal a typical GTPase fold with a P-loop and switch regions critical for nucleotide binding.
