Description

DPP3 encodes dipeptidyl peptidase 3, a zinc-dependent metallopeptidase that cleaves dipeptides from the N-terminus of substrates. It is involved in the degradation of bioactive peptides such as angiotensin III and enkephalins, thereby playing roles in blood pressure regulation and pain modulation. The enzyme is also implicated in oxidative stress response and cancer progression.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Hypertension DPP3 degrades angiotensin III, a vasoconstrictor; altered activity may affect blood pressure regulation. OMIM #126800; NCBI Gene
Pain disorders DPP3 cleaves enkephalins, endogenous opioid peptides; modulation of DPP3 activity influences pain perception. UniProt Q9NY33; PubMed studies
Cancer (various) DPP3 overexpression or mutation linked to tumor growth, metastasis, and poor prognosis in breast, lung, and colorectal cancers. COSMIC; ClinVar; PubMed

Mutation functional classification

Loss of Function (LOF)

Mutations such as p.Met1? and p.Arg163Trp reduce or abolish DPP3 catalytic activity, impairing peptide degradation.

Gain of Function (GOF)

No confirmed gain-of-function mutations reported in DPP3.

Dominant Negative (DN)

No evidence of dominant-negative effects for DPP3 mutations.

Pathways

• Angiotensin III degradation pathway
• Enkephalin degradation pathway
• Oxidative stress response

Protein Summary

DPP3 is a 737-amino-acid zinc-dependent metallopeptidase that removes N-terminal dipeptides from substrates including angiotensin III and enkephalins. It is ubiquitously expressed with highest levels in brain and kidney. The enzyme exists as a monomer and is involved in blood pressure regulation, pain modulation, and cellular redox balance. Altered expression or mutations are associated with hypertension, pain disorders, and several cancers.
