Description

DPP10 encodes a member of the S9B family of serine proteases, but the encoded protein lacks catalytic activity due to mutations in the active site. It functions as a modulator of voltage-gated potassium channels (Kv4.3) by forming a complex with the channel, thereby regulating neuronal excitability and cardiac repolarization. DPP10 is also implicated in asthma susceptibility through its expression in airway epithelial cells and immune modulation.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Asthma DPP10 variants are associated with altered expression in airway epithelium, influencing inflammatory responses and airway hyperresponsiveness. GWAS and linkage studies (OMIM 608209)
Epilepsy DPP10 modulates Kv4.3 channels; dysregulation may contribute to neuronal hyperexcitability. Functional studies (UniProt Q8N608)
Bipolar Disorder Genetic association studies suggest DPP10 polymorphisms may increase risk. Case-control studies (NCBI Gene)

Loss of Function (LOF)

Nonsense mutations (e.g., p.Arg412*, p.Trp189*) lead to truncated, non-functional protein.

Gain of Function (GOF)

Not reported in DPP10.

Dominant Negative (DN)

Not reported in DPP10.

Pathways

• Regulation of cardiac conduction (Kv4.3 channel complex)
• Neuronal system (voltage-gated potassium channels)

Protein Summary

DPP10 is a 796-amino acid type II transmembrane glycoprotein with a large extracellular domain. Despite belonging to the dipeptidyl peptidase family, it lacks catalytic activity due to substitution of the catalytic serine residue. It associates with Kv4.3 potassium channels to modulate their gating properties, influencing action potential duration and neuronal firing. The protein is predominantly expressed in brain and lung tissues.
