Description

DEFA6 encodes defensin alpha 6, a member of the alpha-defensin family of antimicrobial peptides. These peptides are produced by Paneth cells in the small intestine and contribute to innate immunity by disrupting microbial membranes. DEFA6 is specifically expressed in intestinal crypts and plays a role in maintaining gut homeostasis and defense against pathogens.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Crohn's disease Reduced DEFA6 expression may impair antimicrobial defense, contributing to intestinal inflammation. PMID: 19252488
Inflammatory bowel disease Altered defensin levels are associated with dysbiosis and mucosal barrier dysfunction. PMID: 16916644
Colorectal cancer DEFA6 expression is downregulated in some colorectal tumors, potentially affecting tumor microenvironment. PMID: 21536890

Protein Summary

Defensin alpha 6 (DEFA6) is a 32-amino-acid mature peptide (after signal and propeptide cleavage) that forms a triple-stranded beta-sheet structure stabilized by three disulfide bonds. It exhibits broad-spectrum antimicrobial activity against Gram-positive and Gram-negative bacteria, fungi, and some viruses. DEFA6 is stored in Paneth cell granules and secreted into the intestinal lumen, where it contributes to the mucosal barrier.
