Description

DCAF1 (DDB1 and CUL4 Associated Factor 1) encodes a substrate receptor for the CUL4-DDB1 E3 ubiquitin ligase complex. It targets proteins for ubiquitination and proteasomal degradation, playing roles in cell cycle regulation, DNA replication, and chromatin remodeling. DCAF1 is also exploited by HIV-1 Vpr to induce cell cycle arrest and promote viral replication.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Cancer (various types) DCAF1 overexpression or mutation alters ubiquitination of tumor suppressors (e.g., p53, MCM10), promoting genomic instability and proliferation. PMID: 23541922, COSMIC
HIV-1 infection HIV-1 Vpr binds DCAF1 to recruit the CRL4-DCAF1 complex, leading to degradation of host restriction factors and G2/M cell cycle arrest. PMID: 17693256, 17981126

Protein Summary

DCAF1 is a 1507-amino acid protein containing WD40 repeats that mediate substrate recognition. It serves as a substrate receptor for the CUL4-DDB1 ubiquitin ligase complex, targeting proteins such as MCM10 and p53 for degradation. DCAF1 is essential for DNA replication and cell cycle progression, and its interaction with HIV-1 Vpr is critical for viral pathogenesis.
