Description

DAPK3 (death-associated protein kinase 3) encodes a serine/threonine protein kinase that belongs to the DAPK family. The protein is involved in multiple cellular processes including apoptosis, autophagy, and smooth muscle contraction. It acts as a tumor suppressor by promoting cell death and is also a key regulator of actomyosin contractility through phosphorylation of myosin light chain. DAPK3 is widely expressed and its dysregulation is implicated in various cancers and cardiovascular disorders.

Protein Summary

DAPK3 is a 454-amino acid serine/threonine kinase with an N-terminal kinase domain, a leucine zipper domain, and a C-terminal calmodulin-binding region. It phosphorylates myosin light chain and other substrates to regulate actomyosin contractility. The protein is activated by dimerization and autophosphorylation, and it promotes apoptosis through interaction with DAPK1 and other death-associated proteins. DAPK3 is widely expressed in human tissues, with highest levels in smooth and skeletal muscle.
