Description
The CXCL8 gene encodes interleukin-8 (IL-8), a member of the CXC chemokine family. IL-8 is a potent chemoattractant for neutrophils, basophils, and T-cells, and plays a critical role in the inflammatory response, angiogenesis, and tumor progression. It is produced by multiple cell types including macrophages, epithelial cells, and endothelial cells in response to inflammatory stimuli such as TNF-alpha, IL-1, and bacterial products.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Chronic obstructive pulmonary disease (COPD) | Elevated CXCL8 expression recruits neutrophils to airways, contributing to tissue damage and inflammation. | NCBI Gene, OMIM |
| Rheumatoid arthritis | Overexpression of CXCL8 in synovial fluid promotes neutrophil infiltration and joint destruction. | NCBI Gene, OMIM |
| Colorectal cancer | CXCL8 acts as a growth factor and angiogenic factor, promoting tumor progression and metastasis. | COSMIC, NCBI Gene |
| Bronchial asthma | Increased CXCL8 levels correlate with airway neutrophilia and disease severity. | NCBI Gene, OMIM |
| Sepsis | Systemic elevation of CXCL8 contributes to neutrophil-mediated organ damage. | NCBI Gene |

Protein Summary
Interleukin-8 (IL-8) is a 99-amino-acid protein (UniProt P10145) that is secreted as a homodimer. It binds to CXCR1 and CXCR2 receptors on neutrophils, triggering chemotaxis, degranulation, and respiratory burst. IL-8 is also implicated in tumor angiogenesis and metastasis. The protein contains a characteristic CXC motif (Cys-X-Cys) essential for receptor interaction.
