Description
CXCL6 (C-X-C Motif Chemokine Ligand 6) is a chemokine gene encoding a small cytokine belonging to the CXC chemokine family. The encoded protein, granulocyte chemotactic protein 2 (GCP-2), is a potent chemoattractant for neutrophils and plays a key role in inflammatory responses, angiogenesis, and tumor progression. CXCL6 signals through the chemokine receptors CXCR1 and CXCR2.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Inflammatory bowel disease | CXCL6 overexpression recruits neutrophils to intestinal mucosa, exacerbating inflammation | PMID: 21559306 |
| Rheumatoid arthritis | Elevated CXCL6 in synovial fluid promotes neutrophil infiltration and joint destruction | PMID: 12070119 |
| Non-small cell lung cancer | CXCL6 promotes tumor angiogenesis and metastasis via CXCR2 signaling | PMID: 26921324 |
| Colorectal cancer | CXCL6 expression correlates with poor prognosis and promotes epithelial-mesenchymal transition | PMID: 29593339 |

Protein Summary
CXCL6 (GCP-2) is a 114-amino-acid secreted chemokine with a typical CXC motif. It binds to CXCR1 and CXCR2 receptors, mediating neutrophil chemotaxis, degranulation, and angiogenesis. The protein is involved in acute and chronic inflammation, wound healing, and tumor progression. Post-translational processing includes signal peptide cleavage and disulfide bond formation.
