Description

CXCL5 (C-X-C Motif Chemokine Ligand 5) is a chemokine gene that encodes a protein belonging to the CXC chemokine family. This protein is a potent chemoattractant for neutrophils, playing a key role in inflammatory responses and immune regulation. It is secreted by various cell types, including epithelial cells, macrophages, and tumor cells, and exerts its effects by binding to the chemokine receptor CXCR2. CXCL5 is involved in angiogenesis, wound healing, and has been implicated in several inflammatory diseases and cancers.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Colorectal Cancer CXCL5 overexpression promotes tumor growth and metastasis via CXCR2 signaling, enhancing angiogenesis and neutrophil infiltration. PMID: 25605247; COSMIC
Non-Small Cell Lung Cancer Elevated CXCL5 levels correlate with poor prognosis; it recruits neutrophils to the tumor microenvironment, facilitating immune evasion. PMID: 26928204; ClinVar
Rheumatoid Arthritis CXCL5 is upregulated in synovial fluid, attracting neutrophils to joints and contributing to chronic inflammation. PMID: 11590182; OMIM 600324
Chronic Obstructive Pulmonary Disease (COPD) Increased CXCL5 expression in airway epithelium drives neutrophil-mediated inflammation and tissue damage. PMID: 20851989; NCBI Gene

Loss of Function (LOF)

No well-characterized loss-of-function mutations reported in CXCL5.

Gain of Function (GOF)

Overexpression or amplification of CXCL5 is common in cancers, leading to enhanced chemokine signaling and tumor progression.

Dominant Negative (DN)

No dominant-negative mutations have been described for CXCL5.

Protein Summary

The CXCL5 protein, also known as ENA-78, is a 114-amino-acid chemokine with a characteristic CXC motif. It is secreted as a monomer and binds to the G-protein-coupled receptor CXCR2. The protein is involved in neutrophil recruitment, angiogenesis, and tissue remodeling. Its expression is induced by pro-inflammatory cytokines such as IL-1 and TNF-alpha. In cancer, CXCL5 promotes tumor growth and metastasis by modulating the tumor microenvironment.
