Description

CXCL3 (C-X-C Motif Chemokine Ligand 3) is a member of the CXC chemokine family. It encodes a secreted protein that binds to the chemokine receptor CXCR2. CXCL3 functions as a chemoattractant for neutrophils and other immune cells, playing a key role in inflammation, wound healing, and angiogenesis. Overexpression is associated with various cancers and inflammatory diseases.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Inflammatory bowel disease CXCL3 recruits neutrophils to inflamed intestinal mucosa via CXCR2 PMID: 23470731
Colorectal cancer CXCL3 promotes tumor growth and angiogenesis through CXCR2 signaling PMID: 25964219
Breast cancer CXCL3 expression correlates with metastasis and poor prognosis PMID: 27397505
Rheumatoid arthritis CXCL3 mediates neutrophil infiltration into synovial tissue PMID: 12882850
Psoriasis Elevated CXCL3 levels in skin lesions contribute to chronic inflammation PMID: 17641037

Mutation functional classification

Loss of Function (LOF)

No well-characterized loss-of-function mutations reported in CXCL3.

Gain of Function (GOF)

Overexpression in tumors suggests potential gain-of-function via increased chemotaxis and angiogenesis.

Dominant Negative (DN)

Not described for CXCL3.

Protein Summary

CXCL3 is a 107-amino acid secreted chemokine (UniProt P19876) with a typical CXC motif (Cys-X-Cys). It forms homodimers and binds to the G protein-coupled receptor CXCR2. The protein is involved in neutrophil recruitment, angiogenesis, and tumor progression. Post-translational modifications include signal peptide cleavage (residues 1-34) and disulfide bond formation.
