Description

CXCL17 (C-X-C Motif Chemokine Ligand 17) is a secreted chemokine that functions in mucosal immunity, angiogenesis, and tumor biology. It is expressed in mucosal tissues (lung, stomach, colon) and acts as a chemoattractant for monocytes, macrophages, and dendritic cells. CXCL17 is also implicated in tumor progression and metastasis, with overexpression observed in various cancers.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Colorectal Cancer Overexpression of CXCL17 promotes tumor growth and angiogenesis via recruitment of myeloid-derived suppressor cells (MDSCs). PMID: 25934862
Gastric Cancer CXCL17 is upregulated in gastric cancer tissues and correlates with poor prognosis, potentially through immune evasion mechanisms. PMID: 27323851
Lung Cancer CXCL17 expression is elevated in non-small cell lung cancer (NSCLC) and associated with increased microvessel density. PMID: 23149921
Inflammatory Bowel Disease CXCL17 is downregulated in ulcerative colitis mucosa, suggesting a role in mucosal homeostasis. PMID: 21880725

Mutation functional classification

Loss of Function (LOF)

No confirmed loss-of-function mutations reported in CXCL17.

Gain of Function (GOF)

Overexpression in tumors suggests potential gain-of-function in cancer contexts.

Dominant Negative (DN)

No dominant-negative mutations described.

Pathways

• Chemokine signaling pathway (KEGG: hsa04062)
• Cytokine-cytokine receptor interaction (KEGG: hsa04060)

Protein Summary

CXCL17 is a 119-amino acid secreted chemokine containing a C-X-C motif. It is structurally distinct from other chemokines and is highly conserved in mammals. The protein is involved in recruiting immune cells to mucosal surfaces and promoting angiogenesis. Its expression is regulated by inflammatory stimuli and is frequently dysregulated in cancer.
