Description
CSRP1 encodes cysteine and glycine rich protein 1, a member of the CSRP family of LIM domain proteins. It functions as a molecular adapter linking actin cytoskeleton dynamics to gene expression regulation. The protein contains two LIM zinc-binding domains that mediate protein-protein interactions, and is involved in cell proliferation, differentiation, and migration. CSRP1 is widely expressed and plays roles in cardiac development, smooth muscle function, and cancer progression.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Cardiovascular disease | CSRP1 regulates smooth muscle cell differentiation and vascular remodeling; altered expression linked to hypertension and atherosclerosis. | PMID: 14561758 |
| Cancer (multiple types) | CSRP1 overexpression promotes cell migration and invasion in breast, prostate, and colorectal cancers via actin cytoskeleton remodeling. | PMID: 25605246 |
| Hepatocellular carcinoma | CSRP1 upregulation correlates with poor prognosis and metastasis; modulates epithelial-mesenchymal transition. | PMID: 29367602 |

Mutations & Variants
Hotspot Mutations
| Variant | Type | Frequency | Functional Description |
|---|---|---|---|
| c.1A>G (p.Met1?) | Missense | <0.01% | Loss of start codon; likely loss of function |
| c.205C>T (p.Arg69Trp) | Missense | <0.01% | Alters LIM domain; reduced actin binding |
| c.346G>A (p.Gly116Ser) | Missense | <0.01% | Unknown functional effect |
Mutation functional classification
Loss of Function (LOF)
Rare missense variants in LIM domains (e.g., p.Arg69Trp) impair actin binding and nuclear localization.
Gain of Function (GOF)
Not well documented; overexpression in cancer suggests potential gain-of-function via increased cell motility.
Dominant Negative (DN)
No confirmed dominant-negative mutations reported.

Protein Summary
CSRP1 (CRP1) is a 193-amino acid protein with two LIM domains (LIM1 and LIM2) that coordinate zinc ions and mediate interactions with actin, zyxin, and other cytoskeletal proteins. It shuttles between the cytoplasm and nucleus, linking actin dynamics to transcriptional regulation. CRP1 is essential for smooth muscle cell differentiation and cardiac development. In cancer, elevated CRP1 enhances cell migration and invasion, making it a potential biomarker and therapeutic target.
