Description

CSNK2A2 encodes the alpha' catalytic subunit of casein kinase 2 (CK2), a constitutively active serine/threonine protein kinase. CK2 phosphorylates a wide range of substrates involved in cell proliferation, apoptosis, circadian rhythm, and DNA damage response. The alpha' subunit is distinct from the alpha subunit (CSNK2A1) and is essential for embryonic development and hematopoiesis. Dysregulation of CSNK2A2 is implicated in various cancers and neurodegenerative disorders.

Protein Summary

The CSNK2A2 protein (UniProt P19784) is a 350-amino acid catalytic subunit of casein kinase 2 (CK2). It contains an N-terminal kinase domain (residues 1-300) and a C-terminal regulatory region. CK2 is a tetramer composed of two catalytic (alpha or alpha') and two regulatory (beta) subunits. The alpha' subunit is highly conserved and ubiquitously expressed, with highest levels in testis, bone marrow, and lymphoid tissues. It phosphorylates over 300 substrates, including p53, PTEN, and β-catenin, regulating cell survival, proliferation, and DNA repair. CK2 activity is often elevated in cancers, making it a therapeutic target.
