Description
CPA4 (Carboxypeptidase A4) encodes a member of the metallocarboxypeptidase family that cleaves C-terminal aromatic or branched-chain amino acids from proteins. It is secreted as a zymogen and activated by proteolytic cleavage. CPA4 is involved in digestive peptide processing, hormone maturation, and has been implicated in tumor progression, particularly in pancreatic and prostate cancers.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Pancreatic cancer | Overexpression promotes cell invasion and metastasis; may act via ECM remodeling | PMID: 21912567 |
| Prostate cancer | Upregulated in androgen-independent prostate cancer; associated with poor prognosis | PMID: 16397024 |
| Colorectal cancer | Increased expression correlates with lymph node metastasis and advanced stage | PMID: 25642768 |
| Hepatocellular carcinoma | CPA4 overexpression linked to tumor size and vascular invasion | PMID: 28289163 |

Protein Summary
CPA4 is a 421-amino acid secreted metallocarboxypeptidase that requires zinc for catalytic activity. It is synthesized as a preproprotein and activated by removal of the signal peptide and propeptide. The mature enzyme specifically cleaves C-terminal aromatic and branched-chain amino acids. CPA4 is highly expressed in the pancreas and prostate, and its overexpression in several cancers suggests a role in tumor invasion and metastasis.
