Description

The CORT gene encodes cortistatin, a neuropeptide structurally related to somatostatin. It is expressed primarily in the central nervous system and immune cells, where it modulates neurotransmission, sleep regulation, and immune responses. Cortistatin binds to somatostatin receptors (SSTR1-5) and has distinct anti-inflammatory and neuroprotective properties.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Alzheimer's disease Cortistatin levels are altered in brain regions affected by Alzheimer's; potential neuroprotective role through modulation of inflammation and synaptic activity. PMID: 19013282
Inflammatory bowel disease Cortistatin reduces intestinal inflammation by inhibiting pro-inflammatory cytokine release and promoting regulatory T cell activity. PMID: 17086103
Rheumatoid arthritis Cortistatin suppresses synovial inflammation and joint destruction in experimental models via somatostatin receptor activation. PMID: 19155507

Protein Summary

Cortistatin is a 14-29 amino acid neuropeptide (depending on isoform) that shares structural homology with somatostatin. It is produced as a preproprotein and cleaved to generate mature peptides. Cortistatin binds all five somatostatin receptors (SSTR1-5) and exhibits anti-inflammatory, neuroprotective, and sleep-modulating activities. It is predominantly expressed in the brain and immune cells.
