Description

CORIN encodes a transmembrane serine protease that converts pro-atrial natriuretic peptide (pro-ANP) to active ANP, regulating blood pressure and cardiac function. It is expressed primarily in the heart and also in other tissues. Mutations in CORIN are associated with hypertension, preeclampsia, and cardiac hypertrophy.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Hypertension Loss of corin activity reduces ANP activation, impairing natriuresis and vasodilation. PMID: 22923420
Preeclampsia Decreased corin expression in placenta leads to impaired ANP processing and hypertensive complications. PMID: 20884846
Cardiac hypertrophy Reduced corin activity contributes to cardiac remodeling via altered natriuretic peptide signaling. PMID: 22923420

Mutation functional classification

Loss of Function (LOF)

Missense mutations (e.g., p.Thr577Ile) reduce corin's catalytic activity or cell surface expression, impairing ANP activation.

Gain of Function (GOF)

No gain-of-function mutations reported in CORIN.

Dominant Negative (DN)

No dominant-negative mutations reported.

Pathways

• Atrial natriuretic peptide (ANP) signaling pathway
• Regulation of blood pressure by natriuretic peptides

Protein Summary

Corin is a 1042-amino-acid transmembrane serine protease with a cytoplasmic domain, transmembrane region, and an extracellular protease domain. It is synthesized as a zymogen and activated by proteolytic cleavage. Corin specifically cleaves pro-ANP to generate active ANP, which promotes natriuresis, vasodilation, and blood pressure reduction. The protein is expressed on the surface of cardiomyocytes and in other tissues including placenta and kidney.
