Description

CLDN23 (claudin 23) is a protein-coding gene that belongs to the claudin family of tight junction proteins. Claudins are integral membrane proteins that form the paracellular barrier in epithelial and endothelial cell sheets, controlling the flow of molecules between cells. CLDN23 is specifically expressed in the stomach and is involved in gastric epithelial barrier integrity. Altered expression has been linked to gastric cancer and other gastrointestinal disorders.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Gastric cancer Downregulation of CLDN23 disrupts tight junctions, increasing paracellular permeability and promoting tumor invasion. PMID: 16990144; NCBI Gene
Helicobacter pylori infection Reduced CLDN23 expression correlates with H. pylori-induced gastric epithelial barrier dysfunction. PMID: 20026030
Inflammatory bowel disease Altered claudin expression, including CLDN23, contributes to intestinal barrier defects. PMID: 23300010

Protein Summary

Claudin-23 is a 224-amino acid transmembrane protein with four transmembrane domains, two extracellular loops, and intracellular N- and C-termini. It localizes to tight junctions in epithelial cells, particularly in the gastric mucosa. The protein interacts with other claudins to form paracellular pores and seals. Its expression is frequently downregulated in gastric cancer, suggesting a tumor-suppressive role in maintaining epithelial integrity.
