Description

CKAP2 (Cytoskeleton Associated Protein 2) encodes a microtubule-associated protein that localizes to the mitotic spindle and centrosomes. It stabilizes microtubules and is required for proper cell cycle progression, particularly during mitosis. Expression is cell cycle-dependent, peaking in G2/M phase. CKAP2 is overexpressed in various cancers and may contribute to tumorigenesis.

Mutation functional classification

Loss of Function (LOF)

Frameshift and nonsense mutations are predicted to cause loss of function, potentially impairing mitotic progression.

Gain of Function (GOF)

Overexpression in tumors suggests possible gain-of-function in cancer contexts, but specific activating mutations are not well characterized.

Dominant Negative (DN)

No dominant-negative mutations have been reported for CKAP2.

Protein Summary

CKAP2 is a 683-amino acid protein (UniProt Q8WWK9) that binds and stabilizes microtubules. It contains a coiled-coil domain and localizes to the mitotic spindle and centrosomes during mitosis. The protein is phosphorylated during the cell cycle and is degraded after mitosis. CKAP2 interacts with tubulin and other microtubule-associated proteins to regulate spindle dynamics.
