Description

CHTOP (Chromatin Target Of PRMT1) encodes a small nuclear protein that shuttles between the nucleus and cytoplasm. It is involved in chromatin remodeling, mRNA processing, and transcriptional regulation. CHTOP interacts with PRMT1 to modulate histone arginine methylation and is also a component of the TREX complex, facilitating mRNA export. Overexpression and mutations of CHTOP have been implicated in various cancers, including glioblastoma and breast cancer.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Glioblastoma CHTOP overexpression promotes tumor cell proliferation and invasion via altered mRNA processing and PRMT1-mediated chromatin changes. PMID: 23431136, COSMIC
Breast Cancer CHTOP amplification and high expression correlate with poor prognosis; regulates estrogen receptor signaling and cell cycle genes. PMID: 25609607, COSMIC
Colorectal Cancer CHTOP upregulation associated with metastasis and chemoresistance through modulation of Wnt/β-catenin pathway. PMID: 29367642, COSMIC

Protein Summary

CHTOP is a 25 kDa nuclear protein that contains a glycine-arginine-rich (GAR) domain and a C-terminal domain. It binds RNA and interacts with PRMT1, promoting arginine methylation of histones and other proteins. As part of the TREX complex, CHTOP facilitates the export of spliced mRNA from the nucleus to the cytoplasm. Its overexpression in cancers suggests a role in tumor progression by enhancing mRNA processing and export of oncogenic transcripts.
