Description

CDK18 (cyclin dependent kinase 18) is a member of the PCTAIRE subfamily of cyclin-dependent kinases. It encodes a serine/threonine protein kinase that plays a role in cell cycle progression, neuronal differentiation, and DNA damage response. The protein interacts with cyclin A or cyclin E and is involved in regulating centrosome duplication and microtubule organization. CDK18 has been implicated in cancer and neurodegenerative disorders.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast cancer CDK18 overexpression promotes cell proliferation and tumor growth; altered expression linked to poor prognosis COSMIC, ClinVar

Protein Summary

CDK18 is a 473-amino acid serine/threonine kinase with a central catalytic domain and a PCTAIRE motif. It is predominantly expressed in brain and testis. The protein localizes to the cytoplasm and centrosome, and its activity is regulated by binding to cyclin A or cyclin E. CDK18 phosphorylates substrates involved in cell cycle control and cytoskeletal dynamics. Dysregulation of CDK18 is associated with cancer and neurological conditions.
