Description

CDC34 encodes a member of the ubiquitin-conjugating enzyme (E2) family. The encoded protein is required for the G1 to S phase transition of the cell cycle and targets proteins for degradation via the ubiquitin-proteasome pathway. It specifically interacts with the SCF (Skp1-Cul1-F-box) ubiquitin ligase complex to ubiquitinate substrates such as cyclin-dependent kinase inhibitors.

Cancer (multiple types) Dysregulation of CDC34 expression or activity alters ubiquitination of cell cycle regulators, promoting uncontrolled proliferation COSMIC, NCBI
Fanconi Anemia (complementation group) Potential role in DNA repair pathways; altered CDC34 function may impair ubiquitin signaling OMIM, NCBI

Protein Summary

CDC34 is a 236-amino acid ubiquitin-conjugating enzyme (E2) that catalyzes the transfer of ubiquitin from an E1 enzyme to specific substrates in conjunction with an E3 ligase. It plays a critical role in cell cycle progression by targeting cyclin-dependent kinase inhibitors (e.g., p27Kip1) for degradation. The protein contains a conserved active site cysteine (Cys93) required for ubiquitin thioester formation.
