Description

CDC20 encodes a cell cycle protein that acts as a regulatory subunit and activator of the anaphase-promoting complex/cyclosome (APC/C). It is essential for the metaphase-to-anaphase transition by targeting securin and cyclin B1 for degradation. CDC20 also functions in the spindle assembly checkpoint (SAC) by binding to MAD2 and BUBR1. Overexpression is frequently observed in various cancers and correlates with poor prognosis.

Protein Summary

CDC20 is a 55 kDa protein containing seven WD40 repeats that form a beta-propeller structure. It binds to the APC/C via its C-terminal IR tail and activates the complex by recruiting substrates such as securin and cyclin B1. CDC20 is tightly regulated by phosphorylation and by the spindle assembly checkpoint proteins MAD2 and BUBR1. Its overexpression in cancers promotes chromosomal instability and resistance to anti-mitotic drugs.
