Description

CD84 (also known as SLAMF5) is a type I transmembrane glycoprotein belonging to the signaling lymphocytic activation molecule (SLAM) family of immunoreceptors. It is expressed on various hematopoietic cells, including T cells, B cells, NK cells, platelets, and monocytes. CD84 functions as a homophilic adhesion molecule, mediating cell-cell interactions and modulating immune responses. It contains an extracellular domain with immunoglobulin-like domains and an intracellular domain with immunoreceptor tyrosine-based switch motifs (ITSMs) that recruit SH2 domain-containing proteins (e.g., SAP, EAT-2) to transduce signals. CD84 is involved in immune regulation, platelet activation, and has been implicated in autoimmune diseases and cancer.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Systemic Lupus Erythematosus (SLE) CD84 expression is altered on immune cells, contributing to aberrant immune activation and autoantibody production. PMID: 25605269 (NCBI)
Rheumatoid Arthritis CD84 is overexpressed in synovial fluid and tissue, promoting inflammatory cell infiltration and joint destruction. PMID: 25877820 (NCBI)
Chronic Lymphocytic Leukemia (CLL) CD84 is highly expressed on CLL cells, supporting tumor cell survival and microenvironment interactions. PMID: 26582362 (NCBI)
Multiple Myeloma CD84 expression on myeloma cells enhances adhesion to bone marrow stromal cells, contributing to drug resistance. PMID: 27325180 (NCBI)
Infectious Diseases (e.g., HIV) CD84 modulates T cell and NK cell responses, affecting viral control and immune exhaustion. PMID: 29263212 (NCBI)

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
rs3824660 (Intronic variant) SNV Allele frequency ~0.35 (1000 Genomes) Associated with altered CD84 expression in immune cells; linked to SLE susceptibility.
rs2295809 (Missense variant, p.Thr45Ala) SNV Allele frequency ~0.12 (ExAC) May affect protein stability or ligand binding; clinical significance uncertain.
c.123+1G>A (Splice donor variant) Splice site Rare (ClinVar) Predicted to disrupt splicing, leading to loss of function; associated with immunodeficiency in one case report.
p.Arg62* (Nonsense variant) Nonsense Rare (COSMIC) Truncates protein, likely loss of function; observed in a cancer sample.
Mutation functional classification

Loss of Function (LOF)

Loss-of-function mutations (e.g., nonsense, frameshift) in CD84 are rare and may impair immune cell adhesion and signaling, potentially leading to immunodeficiency or altered immune responses.

Gain of Function (GOF)

No clear gain-of-function mutations have been reported for CD84. Overexpression, rather than mutation, is more common in disease contexts.

Dominant Negative (DN)

No dominant-negative mutations have been described for CD84.

Pathways

• SLAM family signaling pathway (SAP/ERS)
• T cell receptor signaling pathway
• B cell receptor signaling pathway
• NK cell mediated cytotoxicity
• Platelet activation

Protein Summary

The CD84 protein is a 339-amino acid type I transmembrane glycoprotein with a molecular weight of ~37 kDa (unglycosylated). It consists of an extracellular region containing two immunoglobulin-like domains (V-type and C2-type), a transmembrane domain, and a cytoplasmic tail with two immunoreceptor tyrosine-based switch motifs (ITSMs). CD84 undergoes homophilic interactions (CD84-CD84) between adjacent cells, mediating cell adhesion. Upon ligand binding, its ITSMs are phosphorylated by Src family kinases, recruiting SH2 domain-containing proteins such as SAP (SH2D1A) and EAT-2 (SH2D1B), which propagate downstream signaling. CD84 is expressed on most hematopoietic cells and plays roles in immune synapse formation, cytokine production, and platelet aggregation. Alternative splicing generates multiple isoforms, some of which may be soluble.
