Description

CD24 encodes a small, heavily glycosylated glycosylphosphatidylinositol (GPI)-anchored cell surface protein. It functions as a ligand for P-selectin (SELP) and plays roles in cell adhesion, immune modulation, and signal transduction. CD24 is expressed on hematopoietic cells, neural cells, and various epithelial tissues. It is frequently upregulated in multiple cancers and is considered a marker of cancer stem cells. CD24 also modulates B-cell development and T-cell costimulation.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast cancer CD24 overexpression promotes tumor cell adhesion to endothelial cells via P-selectin, facilitating metastasis. PMID: 10647931, PMID: 17699752
Colorectal cancer CD24 expression correlates with poor prognosis and increased metastatic potential. PMID: 15604238, PMID: 19137010
Pancreatic cancer CD24 is a marker of cancer stem cells and contributes to tumor initiation and chemoresistance. PMID: 17699752, PMID: 17909063
Ovarian cancer CD24 overexpression enhances peritoneal dissemination and is associated with aggressive disease. PMID: 16951242
Systemic lupus erythematosus (SLE) CD24 polymorphisms are associated with susceptibility to SLE and other autoimmune diseases. PMID: 17909063, OMIM: 600074
Multiple sclerosis CD24 gene variants influence disease risk and progression. PMID: 17909063

Protein Summary

CD24 is a small, heavily glycosylated GPI-anchored cell surface protein. It acts as a ligand for P-selectin, mediating cell adhesion to activated endothelial cells and platelets. CD24 is involved in immune regulation, including B-cell development and T-cell costimulation. It is widely expressed on hematopoietic cells, neural cells, and many epithelial tissues. Overexpression of CD24 is a hallmark of several cancers, where it promotes tumor cell adhesion, invasion, metastasis, and stem cell-like properties. CD24 is also implicated in autoimmune diseases such as systemic lupus erythematosus and multiple sclerosis.
