Description

The CD209 gene encodes the dendritic cell-specific ICAM-3 grabbing non-integrin (DC-SIGN), a C-type lectin receptor expressed on the surface of dendritic cells and macrophages. DC-SIGN recognizes high-mannose carbohydrates on pathogens and endogenous glycoproteins, mediating pathogen capture, antigen presentation, and immune cell adhesion. It is notably exploited by HIV-1 for trans-infection of T cells and is involved in the recognition of multiple viruses, bacteria, and parasites.

Protein Summary

DC-SIGN is a type II transmembrane C-type lectin with a carbohydrate recognition domain (CRD) that binds mannose- and fucose-containing glycans. It forms tetramers on the cell surface, enhancing avidity for multivalent ligands. The protein mediates pathogen capture, antigen processing, and immune synapse formation. Its cytoplasmic tail contains internalization motifs for endocytosis and signaling.
