Description

The CD200 gene encodes a type I membrane glycoprotein belonging to the immunoglobulin superfamily. CD200 is widely expressed on various cell types, including neurons, endothelial cells, and lymphocytes, and functions as an immune checkpoint molecule. It binds to its receptor CD200R on myeloid cells to deliver inhibitory signals that suppress inflammatory responses and maintain immune homeostasis. CD200 is implicated in immune evasion in cancers, autoimmune diseases, and transplant rejection.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Chronic Lymphocytic Leukemia (CLL) CD200 overexpression on tumor cells inhibits anti-tumor immune responses via CD200R engagement ClinVar, COSMIC
Acute Myeloid Leukemia (AML) CD200 expression correlates with poor prognosis and immune suppression ClinVar, COSMIC
Multiple Myeloma CD200 promotes immune evasion and disease progression ClinVar
Rheumatoid Arthritis Altered CD200-CD200R axis contributes to chronic inflammation OMIM
Alzheimer's Disease CD200 downregulation on neurons reduces microglial regulation, promoting neuroinflammation OMIM

Protein Summary

CD200 is a 278-amino acid type I membrane glycoprotein with two immunoglobulin-like domains (IgV and IgC2). It is expressed on the cell surface and functions as a ligand for the CD200 receptor (CD200R) on myeloid cells. Binding of CD200 to CD200R triggers inhibitory signals that suppress macrophage and microglial activation, cytokine production, and T-cell responses. CD200 is involved in immune regulation, neuroprotection, and tumor immune evasion.
