Description

CCNE2 encodes cyclin E2, a regulatory subunit of cyclin-dependent kinase 2 (CDK2). Cyclin E2 binds CDK2 to drive the G1 to S phase transition of the cell cycle. Overexpression or amplification of CCNE2 is frequently observed in several cancers, including breast and ovarian cancer, and is associated with poor prognosis and resistance to CDK4/6 inhibitors.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast cancer CCNE2 amplification/overexpression leads to CDK2 hyperactivation, promoting unscheduled S-phase entry and genomic instability PMID: 15361871; COSMIC
Ovarian cancer CCNE2 amplification drives cell cycle progression and contributes to platinum resistance PMID: 20098429; COSMIC
Endometrial cancer CCNE2 overexpression correlates with aggressive tumor features and poor survival PMID: 23454725

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.1A>G (p.Met1?) missense <0.1% Potential loss of start codon; likely loss of function
c.832C>T (p.Arg278Trp) missense <0.1% Unknown significance; reported in COSMIC
Amplification (8q22.1) copy number gain ~10-15% in breast cancer Increased cyclin E2 expression; oncogenic driver
Mutation functional classification

Loss of Function (LOF)

Rare missense variants (e.g., p.Met1?) may disrupt protein expression or CDK2 binding, but no established loss-of-function phenotype.

Gain of Function (GOF)

Gene amplification and overexpression are the primary gain-of-function mechanisms, leading to increased CDK2 activity and cell cycle progression.

Dominant Negative (DN)

No dominant-negative mutations have been characterized for CCNE2.

Gene Ontology (GO)

• cell cycle • G1/S transition of mitotic cell cycle

Protein Summary

Cyclin E2 is a 404-amino acid protein (molecular weight ~45 kDa) that contains a cyclin N-terminal domain and a cyclin C-terminal domain. It forms a complex with CDK2, and its expression is cell-cycle regulated, peaking at the G1/S boundary. Cyclin E2 is structurally similar to cyclin E1 but encoded by a distinct gene. Its overexpression is oncogenic and linked to chromosomal instability.
