Description
CCNB2 encodes cyclin B2, a member of the cyclin family that regulates the G2/M transition of the cell cycle. Cyclin B2 binds to and activates CDK1 (cyclin-dependent kinase 1), forming the maturation-promoting factor (MPF) essential for mitosis. Unlike cyclin B1, cyclin B2 is predominantly localized to the Golgi apparatus and is involved in Golgi fragmentation during mitosis. CCNB2 expression is cell cycle-dependent, peaking during G2/M phase. Dysregulation of CCNB2 is implicated in various cancers, where overexpression correlates with poor prognosis and tumor progression.

Mutation functional classification
Loss of Function (LOF)
Frameshift and nonsense mutations that truncate the cyclin box domain impair CDK1 binding and kinase activation, leading to G2/M arrest.
Gain of Function (GOF)
Missense mutations that enhance cyclin B2 stability or CDK1 affinity may promote unscheduled mitotic entry and genomic instability.
Dominant Negative (DN)
Mutations that produce a truncated cyclin B2 lacking the destruction box can inhibit normal cyclin B2 degradation, causing mitotic arrest.

Protein Summary
Cyclin B2 is a 398-amino acid protein (UniProt O95067) with a molecular weight of approximately 45 kDa. It contains a cyclin N-terminal domain (residues 42–160) and a cyclin C-terminal domain (residues 161–280) that mediate CDK1 binding and activation. The protein also harbors a destruction box (RxxLxxxxN) near the N-terminus required for ubiquitin-mediated degradation at the end of mitosis. Cyclin B2 localizes to the Golgi apparatus and centrosomes, where it regulates Golgi fragmentation and mitotic spindle assembly. Its expression is tightly regulated at the transcriptional and post-translational levels.
