Description

CCN5 (Cellular Communication Network Factor 5), also known as WISP2, is a member of the CCN family of matricellular proteins. It is involved in regulating cell proliferation, adhesion, migration, and differentiation. CCN5 is frequently downregulated in various cancers and acts as a tumor suppressor by inhibiting Wnt signaling and promoting apoptosis.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast Cancer CCN5 downregulation promotes epithelial-mesenchymal transition and metastasis PMID: 23431173
Colorectal Cancer Loss of CCN5 expression correlates with poor prognosis and increased invasion PMID: 21947069
Pancreatic Cancer CCN5 inhibits tumor growth and angiogenesis via suppression of Wnt/β-catenin signaling PMID: 24316972
Osteoarthritis CCN5 modulates chondrocyte differentiation and matrix remodeling PMID: 25667113

Protein Summary

CCN5 is a 29 kDa secreted matricellular protein composed of four conserved domains: IGFBP, VWC, TSP1, and CT. It modulates cell-matrix interactions and growth factor signaling. CCN5 is considered a tumor suppressor due to its ability to inhibit Wnt/β-catenin signaling and induce apoptosis in cancer cells.
