Description

CCL7 (C-C Motif Chemokine Ligand 7) is a small cytokine belonging to the CC chemokine family. It is produced by various cell types including monocytes, fibroblasts, and endothelial cells. CCL7 binds to chemokine receptors such as CCR1, CCR2, and CCR3, mediating the recruitment of monocytes, macrophages, T cells, and eosinophils to sites of inflammation. It plays roles in immune surveillance, inflammatory diseases, and tumor microenvironment modulation.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Rheumatoid Arthritis CCL7 promotes monocyte and macrophage infiltration into synovial tissue, contributing to joint inflammation and destruction. Increased CCL7 levels found in synovial fluid and serum of RA patients (PMID: 10935631)
Atherosclerosis CCL7 recruits monocytes to arterial walls, promoting foam cell formation and plaque development. CCL7 expression elevated in atherosclerotic lesions (PMID: 11823457)
Breast Cancer CCL7 secreted by tumor cells attracts macrophages and T cells, influencing tumor progression and metastasis. High CCL7 expression correlates with poor prognosis in breast cancer (PMID: 22952447)
Asthma CCL7 recruits eosinophils and Th2 cells to airways, contributing to airway inflammation and hyperresponsiveness. Elevated CCL7 levels in bronchoalveolar lavage fluid of asthmatics (PMID: 10493732)

Protein Summary

CCL7 is a 99-amino-acid secreted chemokine (precursor) that is processed to a mature form of 76 amino acids. It contains four conserved cysteine residues forming two disulfide bonds characteristic of CC chemokines. The protein binds to G-protein-coupled receptors CCR1, CCR2, and CCR3, inducing intracellular calcium flux and integrin activation. CCL7 is involved in monocyte and eosinophil chemotaxis, and its dysregulation is linked to inflammatory diseases and cancer.
