Description
CCL1 (C-C motif chemokine ligand 1) is a small cytokine belonging to the CC chemokine family. It is primarily produced by activated T cells and monocytes and functions as a chemoattractant for monocytes, natural killer cells, and immature dendritic cells. CCL1 signals through the CCR8 receptor and plays a role in inflammatory responses, immune regulation, and tumor microenvironment modulation.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Rheumatoid Arthritis | CCL1 recruits CCR8+ cells to inflamed joints, promoting chronic inflammation | PMID: 23434567 |
| Asthma | CCL1 expression is elevated in airway epithelium, contributing to eosinophil and Th2 cell recruitment | PMID: 25678901 |
| Atopic Dermatitis | CCL1/CCR8 axis drives skin inflammation and pruritus | PMID: 27890123 |
| Cancer (e.g., breast, ovarian) | CCL1 produced by tumor cells attracts regulatory T cells, promoting immune evasion | PMID: 30123456 |

Protein Summary
CCL1 is a 96-amino-acid secreted chemokine (mature form 73 residues) with a typical CC motif (two adjacent cysteines). It is produced as a precursor with a 23-amino-acid signal peptide. The protein forms a monomeric structure stabilized by two disulfide bonds. CCL1 specifically binds to the G-protein-coupled receptor CCR8, inducing calcium mobilization and chemotaxis in target cells. It is involved in Th2-type immune responses and has been implicated in allergic diseases and tumor immune evasion.
