Description
The CAPG gene encodes a member of the gelsolin/villin family of actin-regulatory proteins. This protein caps the barbed ends of actin filaments, preventing addition or loss of actin monomers, thereby regulating actin filament assembly and disassembly. It is involved in cytoskeletal remodeling, cell motility, and phagocytosis, particularly in macrophages.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Cancer (various types) | Altered CAPG expression may promote tumor cell migration and invasion through dysregulation of actin dynamics. | PubMed studies; COSMIC |
| Inflammatory diseases | CAPG modulates macrophage function and actin remodeling, potentially influencing chronic inflammation. | PubMed; UniProt |

Protein Summary
CAPG is a 38 kDa protein that belongs to the gelsolin superfamily. It binds to the barbed ends of actin filaments with high affinity, blocking elongation and depolymerization. The protein is calcium-sensitive and is predominantly expressed in macrophages and other motile cells, where it regulates cytoskeletal dynamics essential for cell migration and phagocytosis.
