Description
CALCB encodes calcitonin-related polypeptide beta, a member of the calcitonin family of peptides. The gene produces a precursor protein that is proteolytically processed to generate the mature neuropeptide CGRP-beta (CGRP-II). CGRP-beta is a potent vasodilator and neuromodulator involved in pain transmission, particularly in migraine pathophysiology. It is expressed primarily in sensory neurons and the central nervous system.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Migraine | CGRP-beta acts as a vasodilator and nociceptive modulator; elevated levels during migraine attacks | ClinVar, OMIM |
| Hypertension | Vasodilatory effects of CGRP-beta may contribute to blood pressure regulation | NCBI Gene, PubMed |
| Osteoarthritis | CGRP-beta involvement in neurogenic inflammation and pain signaling | PubMed |

Protein Summary
The CALCB protein (UniProt P10092) is a 128-amino acid precursor that is cleaved to produce the mature CGRP-beta peptide (37 amino acids). CGRP-beta contains a disulfide bond between Cys2 and Cys7, forming a ring structure essential for receptor binding. It acts via the calcitonin receptor-like receptor (CALCRL) in complex with receptor activity-modifying proteins (RAMPs), primarily RAMP1. The peptide is a potent vasodilator and plays a key role in nociception, neurogenic inflammation, and migraine.
