Description

The CA9 gene encodes carbonic anhydrase 9 (CAIX), a transmembrane zinc metalloenzyme that catalyzes the reversible hydration of carbon dioxide to bicarbonate and protons. CA9 is a major hypoxia-inducible factor (HIF) target gene, strongly upregulated under hypoxic conditions via HIF-1α binding to a hypoxia response element in its promoter. CAIX plays a critical role in pH regulation in the tumor microenvironment, contributing to extracellular acidification and promoting cancer cell survival, invasion, and metastasis. It is widely expressed in various solid tumors, particularly clear cell renal cell carcinoma (ccRCC), where it serves as a diagnostic biomarker and therapeutic target.

Carbonic anhydrase 9 (CAIX) is a 459-amino acid transmembrane protein with an N-terminal extracellular catalytic domain, a single transmembrane helix, and a short intracellular C-terminal tail. It forms homodimers and catalyzes the reversible hydration of CO2 to HCO3- and H+, regulating pH in hypoxic tissues. CAIX is heavily glycosylated and expressed on the cell surface, where it acidifies the extracellular milieu, facilitating tumor invasion and immune evasion. Its expression is tightly controlled by HIF-1α and is a hallmark of clear cell renal cell carcinoma.
