Description

C5AR2 encodes the C5a complement receptor 2, a seven-transmembrane G protein-coupled receptor that binds complement C5a and C5a-desArg. Unlike C5AR1, C5AR2 does not couple to G proteins and may act as a decoy receptor or modulate C5a signaling. It is expressed in various immune cells and tissues, playing roles in inflammation, lipid metabolism, and immune regulation.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Sepsis C5AR2 modulates C5a-mediated inflammatory responses; altered expression may affect sepsis outcome. PMID: 21788451
Asthma C5AR2 expression on airway epithelial cells may influence C5a-induced inflammation. PMID: 24034620
Obesity/Type 2 Diabetes C5AR2 is involved in adipose tissue inflammation and insulin resistance. PMID: 23263627
Cancer C5AR2 expression in tumor microenvironment may modulate immune evasion. PMID: 25605246

Protein Summary

C5AR2 is a 337-amino acid protein with seven transmembrane domains. It shares homology with C5AR1 but lacks the DRY motif required for G protein coupling. C5AR2 binds C5a and C5a-desArg with high affinity and may internalize ligands, acting as a scavenger receptor. It is expressed on neutrophils, macrophages, and other cells, and modulates inflammatory responses.
