Description

C5AR1 (Complement C5a Receptor 1) is a protein-coding gene that encodes a G-protein coupled receptor (GPCR) for the complement component C5a. This receptor mediates chemotaxis, degranulation, and superoxide production in neutrophils and other immune cells. It plays a critical role in inflammatory responses, host defense, and is implicated in sepsis, autoimmune diseases, and tumor progression.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Sepsis C5a-C5AR1 signaling amplifies inflammatory cytokine release and neutrophil activation, contributing to organ failure. PMID: 25947942
Rheumatoid Arthritis C5AR1 activation promotes synovial inflammation and joint destruction via leukocyte recruitment. PMID: 23300472
Cancer (e.g., ovarian, lung) C5AR1 expression on tumor cells and tumor-associated macrophages supports immune evasion and metastasis. PMID: 27626308
Age-related Macular Degeneration C5AR1-mediated inflammation in retinal pigment epithelium contributes to drusen formation and vision loss. PMID: 23377223

Protein Summary

C5AR1 is a 350-amino acid transmembrane GPCR with an extracellular N-terminus and seven transmembrane domains. It binds C5a with high affinity, activating Gαi and Gα16 proteins, leading to calcium mobilization, MAPK activation, and chemotaxis. The receptor is expressed predominantly on myeloid cells and is a therapeutic target for inflammatory diseases.
