Description

BTG1 (B-cell translocation gene 1) is a protein-coding gene that functions as a tumor suppressor. It is a member of the BTG/Tob family of anti-proliferative genes. BTG1 regulates cell cycle progression, differentiation, and apoptosis. It is frequently involved in chromosomal translocations in B-cell malignancies, particularly in acute lymphoblastic leukemia (ALL) and mantle cell lymphoma. The gene encodes a protein that interacts with transcription factors and chromatin remodeling complexes to modulate gene expression.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Acute lymphoblastic leukemia (ALL) Chromosomal translocation t(8;12)(q24;q22) involving BTG1 and MYC leads to dysregulation of cell proliferation. COSMIC, ClinVar
Mantle cell lymphoma BTG1 deletions and mutations contribute to disease pathogenesis by impairing cell cycle control. COSMIC, ClinVar
Diffuse large B-cell lymphoma Somatic mutations in BTG1 are recurrent and associated with poor prognosis. COSMIC, ClinVar

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.1A>G Missense 0.5% p.Met1Val; affects translation initiation
c.200C>T Nonsense 0.3% p.Gln67*; premature stop codon, loss of function
c.300_301insA Frameshift 0.2% p.Leu101Thrfs*5; truncated protein
c.400G>A Missense 0.4% p.Gly134Arg; altered protein function
Mutation functional classification

Loss of Function (LOF)

Nonsense and frameshift mutations (e.g., p.Gln67*, p.Leu101Thrfs*5) result in truncated, non-functional BTG1 protein, impairing its anti-proliferative activity.

Gain of Function (GOF)

No gain-of-function mutations have been reported for BTG1.

Dominant Negative (DN)

Missense mutations (e.g., p.Gly134Arg) may act in a dominant-negative manner by interfering with wild-type BTG1 function.

Pathways

• Cell cycle - G1/S transition
• p53 signaling pathway
• Apoptosis

Protein Summary

The BTG1 protein is a 171-amino acid anti-proliferative factor. It contains a conserved N-terminal domain (BTG domain) that mediates protein-protein interactions. BTG1 localizes to the nucleus and acts as a transcriptional corepressor. It regulates cell cycle progression by inhibiting cyclin D1 expression and promoting p21 expression. BTG1 also interacts with the CCR4-NOT complex to modulate mRNA stability. Post-translational modifications include phosphorylation and ubiquitination, which regulate its stability and function.
