Description

BRPF3 (Bromodomain and PHD Finger Containing 3) encodes a chromatin-associated protein that functions as a scaffold for the HBO1 (KAT7) histone acetyltransferase complex. It contains multiple domains including a bromodomain, two PHD fingers, and a PWWP domain, which mediate binding to acetylated histones and methylated histones, respectively. BRPF3 is a 1,117-amino acid multidomain protein (UniProt Q9Y6K1) that acts as a scaffold for the HBO1 histone acetyltransferase complex. It contains an N-terminal PWWP domain, two PHD fingers, a bromodomain, and a C-terminal domain. The PWWP domain binds H3K36me3, PHD fingers recognize unmodified H3K4 and H3K4me0, and the bromodomain binds acetylated lysines on histones. BRPF3 enhances HBO1 catalytic activity and substrate specificity, promoting acetylation of histone H3 at lysine 14 (H3K14ac) and histone H4 at lysine 5/8/12 (H4K5ac, H4K8ac, H4K12ac).
