Description

BRD3 (Bromodomain Containing 3) is a protein-coding gene that encodes a member of the BET (bromodomain and extra-terminal) family. The protein contains two bromodomains that recognize acetylated lysine residues on histones, facilitating chromatin remodeling and transcriptional activation. BRD3 is involved in cell cycle regulation, gene expression, and has been implicated in cancer, particularly through fusion with NUTM1 in NUT midline carcinoma.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
NUT midline carcinoma BRD3-NUT fusion protein drives oncogenesis by aberrantly activating transcription of pro-proliferative genes PMID: 21575861
Cancer (general) BRD3 overexpression or mutation may contribute to tumorigenesis via dysregulated chromatin binding PMID: 21575861

BRD3-NUT fusion Gene fusion Rare Oncogenic; produces a chimeric protein that drives NUT midline carcinoma
Missense variants Missense Unknown Potential impact on bromodomain function; not well characterized
Mutation functional classification

Loss of Function (LOF)

Not commonly reported; loss of BRD3 function may impair normal transcriptional regulation.

Gain of Function (GOF)

BRD3-NUT fusion acts as a gain-of-function oncoprotein by aberrantly activating transcription.

Dominant Negative (DN)

Not documented for BRD3.

Pathways

• Chromatin organization
• Transcriptional regulation by BET proteins
• Cell cycle

Protein Summary

BRD3 is a 726-amino acid protein belonging to the BET family. It contains two N-terminal bromodomains (BD1 and BD2) that bind acetylated histones, and an extra-terminal (ET) domain involved in protein-protein interactions. BRD3 localizes to the nucleus and regulates transcription by recruiting transcriptional complexes to chromatin. It is ubiquitously expressed with higher levels in testis and hematopoietic tissues.
