Description
BAG4 (BCL2-associated athanogene 4) encodes a member of the BAG family of co-chaperone proteins. The protein contains a BAG domain that interacts with the ATPase domain of Hsp70/Hsc70, modulating chaperone activity. BAG4 also binds to the death domain of tumor necrosis factor receptor 1 (TNFR1) and acts as a silencer of death domain (SODD), thereby inhibiting TNFR1-mediated apoptosis. It is involved in regulating cell survival, proliferation, and stress responses.

Protein Summary
BAG4 is a 457-amino acid protein (UniProt O95429) that functions as a co-chaperone for Hsp70/Hsc70 and as a negative regulator of TNFR1 signaling. It contains a conserved BAG domain at the C-terminus and a death domain-binding region. The protein is widely expressed, with highest levels in testis and lung. BAG4 modulates apoptosis, cell survival, and protein homeostasis. Its dysregulation has been implicated in cancer and neurodegenerative conditions.
