Description

The BAD gene encodes a pro-apoptotic member of the BCL2 family that promotes cell death by binding to and inhibiting anti-apoptotic proteins such as BCL2 and BCL-XL. BAD is a BH3-only protein that acts as a sensor of cellular stress and survival signals. Its activity is regulated by phosphorylation at multiple serine residues, which controls its localization and interaction with 14-3-3 proteins. Dysregulation of BAD contributes to cancer progression and resistance to therapy.

Loss of Function (LOF)

Mutations that prevent BAD phosphorylation or disrupt BH3 domain reduce pro-apoptotic activity, leading to enhanced cell survival.

Gain of Function (GOF)

Not well documented; no common gain-of-function mutations reported in major databases.

Dominant Negative (DN)

Not established for BAD; no dominant-negative variants described in COSMIC or ClinVar.

Protein Summary

BAD (BCL2 Associated Agonist of Cell Death) is a 168-amino acid pro-apoptotic BH3-only protein. It promotes apoptosis by heterodimerizing with anti-apoptotic BCL2 family members (e.g., BCL2, BCL-XL), thereby neutralizing their survival function. BAD activity is tightly regulated by phosphorylation at Ser75, Ser99, and Ser118, which promotes binding to 14-3-3 proteins and cytoplasmic sequestration. Dephosphorylation in response to growth factor withdrawal or cellular stress allows BAD to translocate to mitochondria and initiate apoptosis. BAD is widely expressed in human tissues and is implicated in cancer, where its inactivation contributes to chemoresistance.
