Description

ARPC2 encodes the p34 subunit of the actin-related protein 2/3 (ARP2/3) complex, which is essential for actin nucleation and branching. The ARP2/3 complex mediates the formation of branched actin networks, driving cell motility, endocytosis, and intracellular transport. ARPC2 is ubiquitously expressed and its dysregulation is implicated in cancer invasion and metastasis.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Pancreatic cancer Overexpression of ARPC2 enhances ARP2/3 complex activity, promoting actin polymerization and invasive cell migration PMID: 31209238
Breast cancer ARPC2 upregulation correlates with poor prognosis and increased metastatic potential via lamellipodia formation PMID: 29367642
Colorectal cancer ARPC2 knockdown reduces cell proliferation and invasion in vitro PMID: 30131763

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.437C>T (p.Thr146Met) Missense 0.02% (gnomAD) Unknown functional impact
c.622G>A (p.Glu208Lys) Missense 0.01% (gnomAD) Predicted benign by SIFT
c.1A>G (p.Met1Val) Start loss Rare Likely loss of function
Mutation functional classification

Loss of Function (LOF)

Start loss mutations (e.g., p.Met1Val) are predicted to abolish protein expression, impairing ARP2/3 complex assembly.

Gain of Function (GOF)

No confirmed gain-of-function mutations reported in ARPC2.

Dominant Negative (DN)

No dominant-negative mutations described for ARPC2.

Pathways

Actin nucleation by ARP2/3 complex (Reactome: R-HSA-201688)
Regulation of actin dynamics for phagocytic cup formation (Reactome: R-HSA-2029485)
CDC42 signaling (Reactome: R-HSA-9013423)

Protein Summary

ARPC2 (p34-ARC) is a 34 kDa core subunit of the ARP2/3 complex, which nucleates branched actin filaments. The protein contains a WD40-like domain that mediates interactions with other complex subunits and regulatory factors. ARPC2 is essential for complex stability and localization to the leading edge of migrating cells.
