Description
ARHGDIB (Rho GDP Dissociation Inhibitor Beta) encodes a member of the Rho GDP dissociation inhibitor (RhoGDI) family. The protein regulates the activity of Rho family GTPases by inhibiting the dissociation of GDP from these proteins, thereby maintaining them in an inactive state. It is predominantly expressed in hematopoietic cells and plays a role in cell migration, invasion, and metastasis. Altered expression and mutations of ARHGDIB have been implicated in various cancers and immune disorders.
Disease Associations
| Disease category | Pathophysiological mechanism | Genomic evidence |
|---|---|---|
| Bladder cancer | Reduced expression of ARHGDIB correlates with increased Rho GTPase activity and enhanced metastatic potential. | PMID: 12619160 |
| Breast cancer | Loss of ARHGDIB expression is associated with poor prognosis and increased invasiveness. | PMID: 17699752 |
| Gastric cancer | Downregulation of ARHGDIB promotes cell migration and invasion via RhoA activation. | PMID: 21573173 |
| Lung cancer | ARHGDIB expression is frequently reduced in non-small cell lung cancer, contributing to metastasis. | PMID: 19847194 |
| Ovarian cancer | Low ARHGDIB levels are linked to aggressive tumor behavior and chemoresistance. | PMID: 21750158 |

Protein Summary
ARHGDIB encodes RhoGDI2 (also known as D4GDI), a 201-amino-acid protein that binds to prenylated Rho family GTPases (RhoA, Rac1, Cdc42) and inhibits their activation by preventing GDP release. It is primarily expressed in hematopoietic tissues and acts as a metastasis suppressor in several solid tumors. Loss of ARHGDIB expression is frequently observed in aggressive cancers and correlates with increased Rho GTPase activity, enhanced cell motility, and poor patient outcomes.
