Description

ARHGAP31 encodes a Rho GTPase-activating protein (RhoGAP) that negatively regulates Rho family GTPases, particularly Cdc42 and Rac1. It plays a critical role in cytoskeletal dynamics, cell migration, and development. Mutations in ARHGAP31 are associated with Adams-Oliver syndrome, a rare genetic disorder characterized by scalp defects and limb abnormalities.

Mutation functional classification

Loss of Function (LOF)

Truncating mutations (nonsense) lead to loss of RhoGAP activity, causing Adams-Oliver syndrome.

Gain of Function (GOF)

No gain-of-function mutations reported.

Dominant Negative (DN)

Not described; haploinsufficiency is the proposed mechanism.

Protein Summary

ARHGAP31 encodes a 1449-amino acid protein containing a RhoGAP domain. It inactivates Cdc42 and Rac1 by stimulating their GTP hydrolysis, thereby regulating actin dynamics and cell adhesion. The protein is widely expressed, with highest levels in brain and testis.
