Description

ARHGAP10 encodes a member of the Rho GTPase-activating protein (RhoGAP) family. The protein contains an N-terminal SEC14 domain, a pleckstrin homology (PH) domain, and a C-terminal RhoGAP domain. It functions as a GTPase activator for RhoA and Cdc42, converting them to the inactive GDP-bound state, thereby regulating cytoskeletal reorganization, cell migration, and adhesion.

Loss of Function (LOF)

Frameshift or nonsense mutations that truncate the RhoGAP domain are predicted to impair GTPase activation.

Gain of Function (GOF)

Not well characterized; no recurrent activating mutations reported.

Dominant Negative (DN)

Mutations in the GAP domain may produce dominant-negative effects by sequestering Rho GTPases.

Protein Summary

ARHGAP10 is a 779-amino acid protein that acts as a GTPase-activating protein (GAP) for RhoA and Cdc42. It localizes to the plasma membrane and cytosol, and is involved in regulating actin cytoskeleton dynamics, cell spreading, and focal adhesion turnover. The SEC14 domain may mediate lipid binding.
