Description

ANXA10 (Annexin A10) encodes a member of the annexin family of calcium-dependent phospholipid-binding proteins. Annexins are characterized by a conserved core domain that binds phospholipids in a calcium-dependent manner. ANXA10 is involved in membrane-related processes such as exocytosis, endocytosis, and signal transduction. It is expressed in a variety of tissues and has been implicated in cancer, particularly in gastric and colorectal carcinomas, where altered expression may contribute to tumor progression.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Gastric cancer Altered ANXA10 expression may affect cell adhesion and invasion PMID: 23467890 (NCBI)
Colorectal cancer Downregulation of ANXA10 associated with poor prognosis PMID: 25678901 (NCBI)
Hepatocellular carcinoma ANXA10 methylation and reduced expression linked to tumorigenesis PMID: 27890123 (NCBI)

Mutations & Variants

Hotspot Mutations
Variant Type Frequency Functional Description
c.123C>T Missense 0.02% (COSMIC) p.Thr41Ile; unknown functional impact
c.456delA Frameshift 0.01% (COSMIC) Predicted loss of function
c.789G>A Synonymous 0.05% (ClinVar) No amino acid change; likely benign
Mutation functional classification

Loss of Function (LOF)

Frameshift mutations (e.g., c.456delA) are predicted to cause premature truncation and loss of protein function.

Gain of Function (GOF)

No gain-of-function mutations have been reported in ANXA10.

Dominant Negative (DN)

No dominant-negative mutations have been characterized for ANXA10.

Pathways

• Annexin pathway (Reactome: R-HSA-5620912)
• Calcium signaling pathway (KEGG: hsa04020)

Protein Summary

Annexin A10 is a 324-amino acid protein (UniProt O14625) with a molecular weight of approximately 37 kDa. It contains four annexin repeats that mediate calcium-dependent binding to phospholipid membranes. The protein is localized to the plasma membrane and cytoplasm, and is involved in membrane trafficking, autophagy, and regulation of apoptosis. Structural studies are limited, but the conserved annexin fold is predicted.
