Description

ANXA1 (annexin A1) encodes a calcium-dependent phospholipid-binding protein that belongs to the annexin family. It plays key roles in anti-inflammatory signaling, glucocorticoid-mediated responses, cell proliferation, differentiation, and apoptosis. The protein is a substrate for epidermal growth factor receptor (EGFR) and protein kinase C (PKC). ANXA1 is involved in membrane aggregation, exocytosis, and regulation of cell growth.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Cancer (various types) Dysregulated expression affects cell proliferation, invasion, and metastasis; may act as tumor suppressor or promoter depending on context. Multiple studies in NCBI PubMed; COSMIC database reports mutations in several cancer types.
Inflammatory disorders Anti-inflammatory effects via inhibition of phospholipase A2 and modulation of immune cell migration. ClinVar and OMIM entries; experimental evidence from animal models.
Cardiovascular disease Modulates vascular inflammation and atherosclerosis progression. NCBI Gene and UniProt annotations.

Loss of Function (LOF)

Missense mutations in calcium-binding domains may impair phospholipid binding and anti-inflammatory activity.

Gain of Function (GOF)

Not well characterized; overexpression in some cancers suggests possible oncogenic role.

Protein Summary

Annexin A1 is a 37 kDa protein composed of four annexin repeats, each containing a calcium-binding site. It is predominantly cytosolic but translocates to the plasma membrane upon calcium elevation. The N-terminal domain mediates interactions with S100 proteins and other partners. Post-translational modifications include phosphorylation by EGFR and PKC, and proteolytic cleavage by matrix metalloproteinases. ANXA1 is secreted and acts extracellularly via formyl peptide receptors (FPRs) to exert anti-inflammatory effects.
