Description

AMOTL2 encodes angiomotin-like 2, a member of the angiomotin family of scaffold proteins. It localizes to tight junctions and regulates cell polarity, migration, and proliferation by modulating the Hippo signaling pathway through interaction with YAP1 and WWTR1 (TAZ). AMOTL2 also influences angiogenesis and endothelial cell function.

Disease Associations

Disease category Pathophysiological mechanism Genomic evidence
Breast cancer Dysregulation of AMOTL2 expression alters YAP/TAZ activity, promoting cell proliferation and invasion. COSMIC; PMID: 25944712
Colorectal cancer Reduced AMOTL2 expression correlates with poor prognosis and increased YAP nuclear localization. COSMIC; PMID: 28424480
Hepatocellular carcinoma AMOTL2 overexpression linked to tumor growth via Hippo pathway modulation. COSMIC; PMID: 29395075
Gastric cancer AMOTL2 downregulation associated with metastasis and worse survival. COSMIC; PMID: 27323851

Pathways

• YAP/TAZ regulation (Reactome: R-HSA-8939211)

Protein Summary

AMOTL2 is a 796-amino-acid protein containing coiled-coil domains and a C-terminal PDZ-binding motif. It localizes to tight junctions and interacts with YAP1 and TAZ to inhibit their nuclear translocation, thereby suppressing Hippo target gene transcription. AMOTL2 also binds actin and regulates cytoskeletal dynamics, cell migration, and angiogenesis.
