Description

AIM2 (Absent In Melanoma 2) is a protein-coding gene that encodes a member of the PYHIN (pyrin and HIN domain) protein family. It functions as a cytosolic DNA sensor that recognizes double-stranded DNA from pathogens or damaged host cells, leading to the assembly of the AIM2 inflammasome. This inflammasome activates caspase-1, promoting the maturation of pro-inflammatory cytokines IL-1β and IL-18, and inducing pyroptosis. AIM2 is involved in innate immunity, cancer surveillance, and autoimmune responses.

Protein Summary

The AIM2 protein (UniProt O14862) is 344 amino acids long and contains an N-terminal pyrin domain (PYD) and a C-terminal HIN-200 domain. The HIN domain binds double-stranded DNA, while the PYD domain recruits the adaptor ASC to form the inflammasome complex. This leads to caspase-1 activation, cytokine processing, and pyroptotic cell death. AIM2 is predominantly cytoplasmic and expressed in immune cells and epithelial tissues.
